摘    要:以不同种源水松叶片为材料,应用改良CTAB法提取水松基因组DNA,并对ISSR-PCR反应体系进行优化.结果表明:利用改良CTAB法提取的水松基因组DNA产量高、质量好,符合ISSR分析要求.通过正交试验设计,探讨不同浓度DNA模板、引物、dNPT、Taq DNA聚合酶对水松ISSR-PCR反应体系的影响.

Abstract:The improved CTAB method was used for the extraction of genomic DNA in different populations of Glyptostrobus pensilis,and a satisfactory ISSR reaction system was also established by orthogonal design.The result showed that the improved CTAB method was suitable for the extraction of DNA,and the DNA extracted by this method had higher yield,better quality,and could be used for ISSR-PCR reaction.A number of factors,which affected the results of ISSR-PCR reaction,such as quantity of template DNA,primer,concentration of dNPT and Taq DNA polymerase were studied.
