Agrobacterium-mediated Genetic Transformation of Ipomoea batatas and Re generation of Transgenic Plants

In this paper, the plantlets of sweet potato ( Ipomoea batatas Lam.)c v. Xindazi, obtained by shoot tip culture, were used as test material, the 10 k D zein gene regulated by sporamin promoter was introduced into sweet potato cv . Xi ndazi by Agrobacterium mediated genetic transformation, and some transgenic pla nts were obtained. Excised stems of sweet potato cv. Xindazi were co cultured w ith A.tumefaciens strain LBA 4404 harboring a plasmid pSP10Z, some kanam ycin resistant ( Kan r )buds were formed on the selection medium, which cont ained 75 mg/L kanamycin. The frequency could reach as high as 6.2%. 32.1% of kanamycin resist ant buds rooted on the medium containing 100 mg/L kanamycin, and then formed per fect plants. The results of PCR and PCR Southern analysis confirmed that the 10 kD zein gene was transferred and integrated into the genome of transgenic p lants of sweet potato cv. Xindazi.
