摘    要:以甘薯优良品种"新大紫”的茎尖培养再生植株为试材,经根癌农杆菌介导将甘薯块根贮藏蛋白启动子(Sporaminpromoter)调控下的10kD玉米醇溶蛋白基因导入到"新大紫”中,并获得了转化植株.甘薯品种"新大紫”再生植株的茎切段,与携带有表达载体质粒pSP10Z的根癌农杆菌菌株LBA4404共培养后,在含75mg/L卡那霉素(Kan)的筛选培养基上可

Abstract:In this paper, the plantlets of sweet potato ( Ipomoea batatas Lam.)c v. Xindazi, obtained by shoot tip culture, were used as test material, the 10 k D zein gene regulated by sporamin promoter was introduced into sweet potato cv . Xi ndazi by Agrobacterium mediated genetic transformation, and some transgenic pla nts were obtained. Excised stems of sweet potato cv. Xindazi were co cultured w ith A.tumefaciens strain LBA 4404 harboring a plasmid pSP10Z, some kanam ycin resistant ( Kan r )buds were formed on the selection medium, which cont ained 75 mg/L kanamycin. The frequency could reach as high as 6.2%. 32.1% of kanamycin resist ant buds rooted on the medium containing 100 mg/L kanamycin, and then formed per fect plants. The results of PCR and PCR Southern analysis confirmed that the 10 kD zein gene was transferred and integrated into the genome of transgenic p lants of sweet potato cv. Xindazi.
