纤维素酶法提取资木瓜多糖的工艺研究

摘    要:采用纤维素酶法提取资木瓜多糖,以资木瓜多糖得率为指标,通过单因素和正交试验对影响多糖提取的因素(酶用量、pH值、酶解时间、酶解温度)进行优化。结果表明,酶解温度对资木瓜多糖提取率的影响最大,其次为酶用量和pH值,酶解时间的影响较小。纤维素酶法提取资木瓜多糖的最佳工艺条件为:酶用量1.5%,提取温度55℃,提取时间2.0h,pH值4.0;此条件下资木瓜多糖的产率为9.24%。

Study on Extraction Technology of Chaenomeles speciosa Polysaccharide by Cellulase Method

Abstract:The Chaenomeles speciosa polysaccharide was extracted by cellulase method.With yield rate of polysaccharide as index,the influence factors(enzyme dose,pH value,enzymolysis time and temperatrue)of polysaccharide extraction were optimized through single-factor and orthogonal tests.The results indicated that the influence of enzymolysis temperature on the extraction rate of C.speciosa polysaccharide was greatest,the secondary was enzyme dose and pH value and the influence of enzymolysis time was less.The optimum technological condition for polysaccharide extraction from C.speciosa by cellulase method was as follows:the enzyme dose was 1.5%,the extraction temperature and time were 55 ℃ and 2.0 h and the pH value was 4.0.Under this condition,the yield rate of C.speciosa polysaccharide was 9.24%.
