摘    要:以玉米自交系丹黄25为试材,用农杆菌介导法将玉米淀粉分支酶SBEIIb基因转入玉米中,探讨了农杆菌介导玉米愈伤组织转化的影响因素.结果表明:愈伤组织经过9d的继代,菌液中乙酰丁香酮(AS)浓度为10 mg/L,侵染时间为20 min,共培养60 h为最佳转化条件.获得的4株再生植株经PCR分析鉴定,其中3株表现阳性,证...

Abstract:The SBEIIb gene was transferred into maize inbred line Danhuang 25 by Agrobacterium tumefaciens mediated transformation.Factors influenced transformation efficiency was studied.The results showed that 9 days with subculture of callus,AS concentration in the bacilli concentration at 10 mg/L,20 minutes infection time and co-culture 60 hours were the optimal conditions.PCR analysis on four resistance plants demonstrated that three of them were positive and the foreign gene was integrated into maize genome.
