甘蔗茎部内生细菌的分离及分子鉴定

摘    要:采用稀释涂板法对广东省广州市白云区国家农业科技园区内的甘蔗茎部内生菌进行分离,同时进行16S rDNA扩增检测及同源性分析。结果表明:共分离获得20株不同的内生菌,所获得的内生菌分属于6个类群(Actinobacteria、Alphaproteobacteria、Gammaproteobacteria、Betaproteobacteria、Firmicutes、Acidobacteriales)、16个属(Staphyococcus,Leifsonia、Kineococcus、Frondihabitans、Curtobacterium、Microbacterium、Amnibacterium、Methylobacterium、Rhizobium、Terriglobus、Sphingomonas、Acidisoma、Stenotrophomonas、Enterobacter、Burholderia、Variovorax),其中,Actinobacteria和Alphaproteobacteria类群占总数的65%。除SC-4、SC-11与其同源细菌相似性分别为97%、96%外,其余分离菌与同源菌相似性均在98%以上。本研究结果在一定程度上反映了甘蔗茎部内生菌具有较高的多样性。

Abstract:Based on the dilution method and 16S rDNA, the endophytic bacteria in sugarcane(Saccharum officinarum)stem were isolated and identified, which would provide important reference for the study of the diversity of sugarcane endophytic bacteria and interaction between endophytic bacteria and host. The result showed that 20 isolates obtained in this study were belonged to 6 groups(Actinobacteria, Alphaproteobacteria, Gammaproteobacteria, Betaproteobacteria, Firmicutes, Acidobacteriales), 16 genus(Staphyococcus, Leifsonia, Kineococcus, Frondihabitans, Curtobacterium, Microbacterium, Amnibacterium, Methylobacterium, Rhizobium, Sphingomonas, Acidisoma, Stenotrophomonas, Enterobacter, Burholderia, Variovorax). The homology of most isolates was more than 98% except SC-4 and SC-11, which was only 97% and 96% respectively. It was indicated that sugarcane stem was high diversity of endophytic bacteria to some extent.
