Abstract:The technique of chromosome microdissection and microcloning has been becoming an efficient tool for constructing chromosome or chromosome segment specific DNA libraries since the first successful cloning of specific chromosome regions in 1981. Now, it is widely used in the studies of genetics, medicine, evolution and other fields. In this paper, the development and research progress of this technique in plant are reviewed, including chromosome preparation, chromosome identification, microdissection, microcloning, confirmation of the clones and characterization of chromosome DNA libraries. The application prospects of this technique in plant are also discussed.
