银杏细胞对抗生素的耐受性研究

摘    要:用MS培养基诱导银杏胚产生愈伤组织.分别在MS培养基中加入不同浓度的卡那霉素、潮霉素和杀稻瘟菌素,将银杏愈伤组织转接到含抗菌素的培养基上继续培养11d之后得到以下结果:MS培养基中含潮霉素25mg/L时,银杏愈伤组织变成褐色,停止生长;MS培养基中含同样浓度的杀稻瘟菌素时,银杏愈伤组织变成浅褐色生长变慢;MS培养基中合同样浓度的卡那霉素时,银杏愈伤组织生长良好.银杏细胞对潮霉素的耐受性很弱,可以作为筛选转基因银杏树种的抗生素使用。

Abstract:The results obtained by having transplanted the callus of Ginkgo biloba produced from embryo on normal MS medium into mediums with different antibiotics for 11 days were as follows: (1)the callus cells turned brown and stopped growth on MS medium with 25 mg/L of hygromycin; (2) the callus cells turned light brown and partially stopped growth on MS medium with 25 mg/L blasticedin; (3) the callus cells of Ginkgo biloba grew well on MS medium containing 25 mg/L kanamycin. It showed that the callus cells of Ginkgo biloba were susceptible to hygromycin, and hygromycin could be used as antibiotics for screening of gene transferred varieties of Ginkgo biloba.
