笃斯越橘再生体系的建立

摘    要:以笃斯越橘休眠期的茎段和单芽为外植体,采用改良WPM为基本培养基,研究不同激素、外植体类型对植株再生和生根情况的影响.结果表明,枝条经过室内水培及10℃预处理,以单芽为外植体比以茎段为外植体可更有效快速地获得无菌苗.茎段及单芽的离体增殖生长的最适宜培养基为:WPM+1.5 mg·L-1 ZT+0.3 mg·L-1 NAA;生根最适宜培养基为:1/2WPM+1.5 mg·L-1 IAA+0.1 mg·L-1 IBA,生根成活率达82.3%.

Abstract:The stem segments and single buds of Vaccinium ulignosum in dormancy were used as explants for experiment.Modified WPM was applied to basic medium.The effects of the different hormones and explants type to plant regeneration and rooting were studied.The result showed that the branches were pretreated by hydroponic indoor and kept at temperature 10 ℃,the single buds for explants can effectively get sterile plants than stem segments.The optimal culture medium for stem segments and single buds was WPM+1.5 mg·L-1 ZT+0.3 mg·L-1 NAA;The optimal culture medium for rooting was 1/2 WPM+1.5 mg·L-1 IAA+0.1 mg·L-1 IBA with rooting rate of 82.3%.
