产氨短杆菌产辅酶A反应体系的优化设计

摘    要:采用产氨短杆菌体对Coenzyme A(CoA)酶促合成的最优合成条件进行了研究。经过计算机模拟寻优,得到反应液的最优配方为KH2PO4600mg、氯化十六烷吡啶10mg、泛酸钙70mg、MgSO4·7H2O60mg、半胱氨酸盐酸盐130mg,加水定容至100mL(pH6.0)。CoA的最高效价值为29.99×10^3单位/L,比原配方的效价值提高了2.68倍。

Abstract:The best biosynthesis conditions of the Coenzyme A were studied using Brevibacterium ammoniagenes. Through the computer optimization, the best design is as follows: KH2PO4 600mg, Cetylpyridinium Chloride, Monohydrate (CPC) 10mg,Calcium Pantothenate 70 mg, MgSO4·7H2O60mg, Cysteine Hydrochloride (Anhyarous) 130mg, and water was used to dilute the final volume up to 100 mL (pH 6.0) . The production of Coenzyme A achieved 29.99×10^3, which was 3.68 times the original formula.
