鹅神经肽Y基因的克隆及生物信息学分析

摘    要:克隆出鹅NPY基因并对序列进行分析。根据鸡NPY基因核苷酸序列设计引物,利用RT-PCR方法获得了鹅NPY基因的编码序列,并用相关软件及在线分析方法对其核苷酸及氨基酸序列进行分析。结果表明,鹅NPY基因长度为371 bp,包含294 bp整个开放阅读框,共编码97个氨基酸,其分子量预测值为11.083 kD,等电点为(pI)为5.76。鹅NPY基因核苷酸序列与禽类和哺乳动物的序列同源性分别为96%~97%、86%~88%。氨基酸进化树分析发现NPY基因具有种间多样性,鹅NPY与其他禽类处于同一分支。成功克隆鹅NPY基因CDS序列并对其进行了生物信息学分析,为深入研究其功能奠定基础。

Abstract:This study was to clone and analyze NPY gene of goose. Primers were designed according to the chicken NPY gene nucleic sequence. Goose NPY gene was cloned by RT-PCR. The nucleic sequence of goose NPY gene and the amino acid sequence deduced from the gene were analyzed by related softwares and on-line tools. The results showed that the amplified goose NPY was 371 bp in length, containing one open-reading frame of 294 bp and encoding 97 amino acid residues. The alignment of goose NPY gene cDNA sequence with other species showed the sequence similarities were respectively 96%-97% and 86%-88%. Phylogenetic tree showed NPY gene had varieties among species, and goose NPY was the same with other avian in the cladogram topological structure. Goose NPY was cloned and analyzed in this study, and it would lay a foundation for the further study on the function of NPY gene.
