怀地黄叶片愈伤组织的诱导形成及其可溶性糖含量变化的研究

对怀地黄叶片愈伤组织诱导形成及其可溶性糖含量变化的研究结果表明:1/2MS+6-BA0.5mg/L培养基有利于怀地黄块茎诱导形成无菌苗。单独使用2,4-D有利于叶片愈伤组织的诱导,但不利于愈伤组织的继代培养;2,4-D和6-BA配合使用时,对愈伤组织的增殖具有良好的促进作用,当2,4-D浓度为1.0mg/L时,随6-BA浓度的增加,愈伤组织的生成量也增加,其中,以MS+2,4-D1.0mg/L+6-BA0.4mg/L培养基诱导的愈伤组织的量为最大,且呈黄色、疏松、颗粒状。愈伤组织诱导形成期间可溶性糖含量在不同培养基中均呈现先下降后上升,达到峰值后再下降的趋势。

Studies on the Callus Inducement of Leaf of Rehmannia glutinosa Libosch and Its Variance of Soluble Sugar Content

Studies on the effects of different hormone level combinations on callus induction of Rehmannia glutinosa Libosch. Showed that 1/2 MS 6-BA 0.5mg/L is advantageous to induct aseptic seeding from tuber. It is most favourable for the inducement of aseptic seeding from tuber when using 2,4-D only, but it doesn't fit for the later growth of callus. When 2,4-D mixed with 6-BA, the calli can grows better, and when 2,4-D is at 1.0 mg/L the quantity of calli is increasing with the increasing of 6-BA concentration. MS 2,4-D 1.0mg/L 6-BA 0.4mg/L is the best medium for the inducement of callus. The soluble sugar content in different medium decreases first and then increases, and it decreases again when it gets the maximum.
