摘    要:[目的]纯化培养食蟹猴支持细胞,并测定不同血清浓度对细胞增殖的影响。[方法]分离并纯化食蟹猴支持细胞后,分别以5%、10%、15%、20%4种不同血清浓度的培养基对食蟹猴支持细胞进行培养。利用MTT比色法判断支持细胞的增殖情况,从而确定最佳的血清培养浓度。[结果]5%血清浓度的培养基对细胞增殖能力的影响与10%血清浓度的培养基差异不显著,15%与20%的血清浓度的培养基对细胞增殖能力的影响差异不显著,5%、10%与15%、20%的血清浓度的培养基对细胞增殖能力的影响差异极显著。[结论]15%的血清浓度是支持细胞增殖的最佳比例。

Isolation and Purification of Sertoli Cells in Macaca fascicularis and Effect of Different Concentrations of Serum on Its Proliferation

Abstract:[Objective] To isolate and culture the sertoli cell in in Macaca fascicularis,and determine the effect of different concentrations of serum on its proliferation.[Method] The sertoli cells were firstly isolated and purified,and then cultured respectively in the medium with differentserum concentrations of 5%,10%,15% and 20%.MTT colorimetric method was adopted to detect the proliferation of the sertoil cells so as to determine the optimal serum concentration.[Result] The proliferation capacity of sertoli cells cultured in 5% serum showed no significant difference from that cultured in 10% serum,the same of 15% and 20% serum,while the proliferation capacity of sertoli cells cultured in 5% and 10% serum differed largely from that cultured in 15% and 20% serum.[Conclusion] 15% serum concentration was the best for the proliferation of sertoli cells.
