摘    要:以树型金银花茎尖为材料,比较材料的不同采集时间、预处理方法、解离时间和染色方法对树形金银花茎尖染色体制片的影响。结果表明,取材时间为上午10:00-11:00、预处理方式为饱和对二氯苯处理3~4 h、解离采用1mol/L HCl在60℃下13min、染色液以改良石碳酸品红染色10min染色体制片效果最佳。核型分析结果表明:染色体核型公式为2n=18=8m+8sm+2st,其中第9对染色体具随体,染色体相对长度组成为2n=18=4L+8M2+4M1+2S,属于"2B"型,核型不对称指数为64.47%,属于由对称向不对称演化的过渡型,表明树型金银花是较野生型金银花进化类型。

Abstract:It was compared that the effects of different sampling time,the methods of pretreatments,dissociations time and dyeing time on chromosome preparation of tree Lonicera japonica shoot tips.The results showed that the best experimental protocol for karyotype analysis of tree Lonicera japonica.is better when sampling time is between 10:00AM and 11:00AM,the pretreatment time with saturated p-dichlorobenzene lasts 3-4h,the dissociation with 1mol/L HCl is at 60℃ for 13min,and the dyeing time lasts 10min with modified carbolic acid-magenta.Simultaneously, the karyotype analysis also showed that the karyotype formulae of tree Lonicera japonica is 2n=18= 8m +8sm+2st,where one satellite was found in the ninth chromosomes, the relative length of the chromosome composition was 2n=18=4L+8M2+4M1+2S, belonging to "2B" type.Its index of Asymmetric was 64.47%, and the karyotype asymmetry belongs to transition type from symmetry type to asymmetry one, suggesting that tree Lonicera japonica is relatively evolutionary form compared to wild type Lonicera japonica.
