铁皮石斛组培技术研究

摘    要:目的]为铁皮石斛的产业化发展提供依据。方法]以铁皮石斛组培幼苗为试材,接种于不同培养基和培养条件下,培养60 d后分别取出全部小苗称重;组培苗移栽后统计成活率。结果]4种不同基本培养基中,以N6对铁皮石斛幼苗的增殖效果最好,增产率达1619.0%,MS最差;以椰子汁为附加物时,对铁皮石斛苗的增殖促进作用最大,香蕉泥和马铃薯泥的增殖效果相近;日光灯照下的铁皮石斛苗较高,自然光照下培养的铁皮石斛苗粗壮,叶色浓绿,增殖率高。当苗高为(6.0±1.0)cm时,经1周炼苗后,洗净根部培养基移栽至石灰石碎石滤水层5 cm+锯末8 cm+活苔藓2 cm的基质,成活率在95%以上,植株生长良好。结论]得到了优质、高效、低成本、规模化培育铁皮石斛组培苗的方法。

Studies on Industrialization of Tissue Culture of Dendrobium candidum
Abstract:Objective] The aim was to provide the reference for the industrialization development of Dendrobium candidum.Method] With D.candidum seedlings as test materials,they were inoculated on the different media.After cultivation for 60 d,the seedlings were all taken out to weight and the survival rate was calculated after the tissue culture seedlings were transplanted.Result] Among the 4 kinds of basal media,the proliferation effect of N6 on the seedlings was best with the yield-increasing rate of 1 619.0% and that of MS was worst.With the coconut milk as the additive,its proliferation effect on D.candidum seedling was biggest and the proliferation effect of banana mud and mashed potatoes was similar.The height of seedlings irradiated by fluorescent lamp was higher and the seedlings cultured under the sunlight were stronger with deep green leaves and high proliferation rate.When D.candidum seedlings had height of(6.0±1.0) cm,after hardening-seedling and washing the medium in root,they were transplanted in the matrix of 5 cm limestone spall filtrating layer +8 cm sawdust +2 cm living lichen and their survival rate was over 95% with good growth.Conclusion] The method for industrially culturing D.candidum seedlings with good quality,higher efficiency and low cost was obtained.
