酶解鸡蛋清小(寡)肽混合物对小鼠腹腔巨噬细胞一氧化氮产生的影响

摘    要:研究通过体外试验考察了5种酶解鸡蛋清小肽混合物(MSEE)和5种酶解鸡蛋清寡肽混合物(MOEE)对小鼠腹腔巨噬细胞一氧化氮(NO)产生的影响。结果表明:无论细胞培养介质中是否含LPS,所有MSEE和MOEE均不同程度地提高小鼠腹腔巨噬细胞培养液中NO水平,但不同剂量间和不同小(寡)肽混合物种类间有差异,其中以1.0mg/mL剂量组NO水平最高,且NO水平高低顺序为MSFL>MSPA>MSTR>MSAL>MSPE>对照和MOFL>MOTR>MOPA>MOAL>MOPE>对照。此外还发现,LPS显著影响MSEE和MOEE促小鼠腹腔巨噬细胞NO产生,与不添加LPS组相比,LPS攻毒的MSAL、MSFL、MSPA、MSPE和MSTR组NO水平分别提高了45.1%、69.0%、58.1%、37.9%和61.8%,含LPS的MOAL、MOFL、MOPA、MOPE和MOTR组NO水平分别提高了40.9%、62.9%、49.6%、26.7%和54.4%。这些结果表明,所有MSEE和MOEE(特别是MSFL和MOFL)均能刺激巨噬细胞NO产生,尤其是培养介质中含LPS时NO的产生效应更为明显。

Effect of Mixtures of Small/Oligo-peptides from Enzymic Hydrolysates of Egg White on NO Production in Mice Peritoneal Macrophage in vitro

Abstract:In the present study,we investigated the effect of five different mixtures of small peptides(MSEE) and five different mixtures of oligopeptides(MOEE) from enzymic hydrolysates of egg white on nitric oxides(NO) production in mice peritoneal macrophage in vitro.The results showed that regardless of LPS challenge,all of the MSEEs and MOEEs improved the NO level in supernates of mice peritoneal macrophage,while the difference was observed with NO level by different doses of MSEE or MOEE and different small/oligo-peptide mixtures treatment,respectively.NO level had the obviously response to MSEE and MOEE at the dose of 1.0 mg/mL,the stimulation ability from the highest to the lowest was MSFL>MSPA>MSTR>MSAL>MSPE>control and MOFL>MOTR>MOPA>MOAL>MOPE>control,respectively.Moreover,the results also showed that MSEEs and MOEEs could significantly elevate NO level with the challenge of LPS.Compared with no LPS challenge,the NO levels of MSAL,MSFL,MSPA,MSPE,and MSTR treatment in LPS challenge group were increased by 45.1%,69.0%,58.1%,37.9%,and 61.8%,respectively.The NO levels of MOAL,MOFL,MOPA,MOPE,and MOTR treatment in LPS challenge group were increased by 40.9%,62.9%,49.6%,26.7%,and 54.4%,respectively.Our results suggested that all of MSEEs and MOEEs(particularly MSFL and MOFL) could stimulate NO production in mice peritoneal macrophage in vitro,especially under the challenge of LPS.
