苜蓿属种质资源遗传多样性RAPD分析

摘    要:本文用RAPD标记分析10份苜蓿属(Medicago L.)种质的遗传多样性和亲缘关系,为苜蓿优秀种质资源开发保护和杂交育种提供基础信息。结果表明:33个引物在这10份材料间共扩增出353条带,其中263条带为多态性带,多态位点百分率为74.5%,各引物扩增的条带数为5~17条不等;遗传距离范围为:0.488~0.686,平均值0.592。聚类分析结果表明:10份材料大体划分为3类:草原1号、草原3号、美国苜蓿王和达菲成为第1类;雅酷、呼盟、蒙古和锡盟是第2类;秋柳独自成为1类。

Abstract:The genetic diversity and relationships of the 10 different Medicago L. germplasms were analyzed using Random Amplified Polymorphic DNA (RAPD) markers in order to provide essential information for further exploitation and hybridization. RAPD analysis demonstrated that 33 random primers produced 353 loci among which 263 were polymorphic loci, the percentage of polymorphic loci was 74.5%, and each primer produced 5~17 belts. Genetic similarity changed from 0.488 to 0.686, averaged 0.592. According to clustering analysis, we classified the 10 materials into three groups: CaoYuan Ⅰ, CaoYuan Ⅲ, Alfaking, and Daviluya were the first group, L.Yakuskayayellow, HuMeng, MengGu and XiMeng the second group, and Culuskaya the third group.
