槟榔江水牛群体遗传结构的RAPD分析

摘    要: 为探讨RAPD标记在水牛遗传多样性方面检测的应用价值,以及了解槟榔江水牛的群体遗传结构状况,研究采用随机扩增多态性DNA标记技术对槟榔江水牛20个个体进行了遗传变异分析,从70个随机引物中筛选出15个多态性丰富的引物对其所有个体的总基因组DNA进行了PCR扩增,结果15条引物共产生105种扩增片段,多态片段95条,平均每条引物的扩增带数为7,各引物多态性片段范围在2~12之间,多态频率在40%~100%之间,多态座位平均为90.48%。表明槟榔江水牛的遗传多样性较丰富。

Abstract:In order to estimate the value of the RAPD marker on the study of water buffalo population genetic diversity,as well as the genetic construction on Binglangjiang water buffalo population,the random amplified polymorphic DNA(RAPD) marker was used to analyze genetic variation of 20 Binglangjiang water buffaloes in this paper.Individual genome DNA was amplified with 15 polymorphism random primers which were screened from 70 random primers.105 bands were obtained,of which 95 bands were polymorphic,each primer had 7 bands on average and the fragment number is in between 2~12,frequencies of polymorphic bands is in between 40%~100%,the mean value of polymorphic loci was 90.48%. The results indicated that the genetic diversity of Binglangjiang water buffalo is rich.
