PVP对烟草基因组DNA提取的影响

摘    要:为克服烟叶组织中蛋白质、糖类、色素、烟碱及酚类等物质对烟草基因组DNA提取的干扰,防止DNA产生褐变,获得质量高且适于SRAP-PCR反应需要的DNA,采用改良CTAB法探讨了在提取液中添加不同浓度PVP对烟草基因组DNA提取质量的影响.结果表明:在1%、2%、4%PVP浓度下提取烟草DNA样品的OD260/OD280值均在1.7~1.9之间,OD260/OD280值随着提取液中PVP浓度的提高而增大,DNA褐化程度明显降低,获得的DNA能满足SRAP分析要求.
