摘    要:A-蛋白和羊抗兔血清应用于免疫电镜,检测马铃薯X病毒(PVX)粗汁液,灵敏度比诱捕修饰法高;而且加羊抗兔血清后,病毒粒体比诱捕修饰法明显加粗,在电镜放大2500倍时,就能清晰地观察到。用诱捕双修饰法检测PVX,证实在寄主植物的叶、茎和根中均存在,以叶内含量最高。接种在普通烟上第三天,接种叶就能检测到PVX,接种15天后,寄主体内病毒能达到较高浓度,而且高浓度一直可保持2个月以上;PVX在不同寄主中的含量不同,以普通烟中病毒浓度最高,心叶烟、番茄和黄花烟中次之;昆诺阿藜、千日红和大椒中含毒量极抵。用诱捕双修饰法检测马铃薯薯块休眠芽中的病毒获得成功。

Abstract:The making use of protein A and antirabbit serum of sheep in immune electron microscopy for detecting potato virus X extracts of infected leaves was found of a higher sensitivity than the trapping-decoration method. A further coating of the virus specific antiserum by antirabbit serum of sheep resulted in a considerable increase of the diameters of virus particles and gave a clear-cut observation in electron microscopy field at lower magnification(2500x).Studies on potato virus X by immunoelectron microscopy(trappingdouble decoration method) has proved that potato virus X existed not.only in leaves, but also in stems and roots of host plant, the leaves contained the largest amount of virus though.The virus particles of infected leaves were found 3 days after infection. The virus concentration in host increased after 15 days and the level remained for more than two months.The concentrations of potato virus X are different in various host plants. The highest was found in Nicotiana tabacum "Whit Burley", and higher levels were found in N. glutinosa, Lycopersicon esculentum and N. rustica. It was very low in Chenopodium quinoa, Gomphorena globosa and Capsicum anmuum.The trapping-double decoration method has been applied successfully to detect potato virus in extracts of infected potato dormancy bud.
