潮霉素对紫花苜蓿愈伤组织诱导和分化的影响

摘    要:潮霉素是当今基因工程中的一种转化选择剂。通过对苜蓿叶片诱导阶段、分化阶段施加不同浓度的潮霉素,确定潮霉素对紫花苜蓿叶片愈伤组织形成及其分化的影响。研究表明,潮霉素对紫花苜蓿叶片愈伤组织诱导与分化有显著的抑制作用。当潮霉素浓度达到15mg/L时苜蓿叶片完全无法诱导愈伤;愈伤和分化阶段都添加10mg/L潮霉素时,虽然可以诱导出愈伤组织但无法分化。因此建议在遗传转化时诱导愈伤阶段及分化阶段都添加10mg/L潮霉素。

Abstract:Hygromycin is a selected marker for genetic manipulation. Effect of hygromycin on callus formation and differentiation of M. sativa leaves was studied. The results were shown that inducement and differentiation of M. sativa leaves was inhibited by hygromyein. When the concentrations of hygremycin was up to 15 mg/L, eaUus was not induced. The callus was not differentiated when the concen- tration of hygromyein was up to 10 mg/L in induction and differentiation phase. Consequently,We recommended that the concentrations of hygromyein should be 10 mg/L in induction and differentiation phase.
