珠眉海棠cDNA-AFLP分析体系的建立

摘    要:以珠眉海棠(Malus zumiMats)为研究材料,对cDNA-AFLP反应体系的关键因素(酶切时间、预扩增体系、选择性扩增体系)进行探索,建立了重复性好的cDNA-AFLP分析体系。结果表明,EcoRI和MseI的组合4h可将ds-cDNA酶切完全,与相应接头连接过夜后产物直接用于预扩增,1μl稀释20倍的预扩增产物作为模板用于选择性扩增,可以获得带型丰富且重复性好的结果。珠眉海棠cDNA-AFLP反应体系的建立为进一步的分子生物学研究建立了平台。

Abstract:The cDNA Amplified Fragment Length Polymorphism(cDNA-AFLP) system of Malus zumi Mats was established.The key factors,including enzyme digestion,preamplification and selective amplification,were optimized.The results indicated that double-stranded cDNA of Malus zumi Mats was digested completely after 4h digestion by EcoRI and MseI.The reducible result was obtained when the ligation products without dilution were used as templates for preamplification and 1μl of 20 times-diluted preamplification products for selective amplification.The cDNA-AFLP system was established successfully,which provided platform for further molecular study and gene cloning of Malus zumi Mats.
