甘草下胚轴离体再生体系的建立

摘    要:以甘草下胚轴为外植体,对影响下胚轴离体再生不定芽的因素进行研究。结果表明:KT具有显著作用,与6-BAI、BA结合,可高效诱导下胚轴直接产生不定芽,优化的分化培养基为MS 6-BA 2.0 mg/L KT 0.5 mg/L IBA 0.5 mg/L,芽苗分化率平均为41.5%。同时,进一步研究了不同基因型、预培养处理对甘草下胚轴再生的影响,结果发现:胀果甘草下胚轴离体再生频率最高,达44.7%;预培养4 d对甘草下胚轴再生不定芽具有促进作用。建立甘草下胚轴离体再生不定芽体系,为甘草基因工程品种改良及优良品种快速繁殖打下基础。

Abstract:Factors influencing adventitious bud regeneration of Glycyrrhiza hypocotyls were studied.The results indicated that KT had a prominent effect.Combined with 6-BA and IBA,adventitious bud can be directly induced from hypocotyls of Glycyrrhiza,the polarization medium was MS 6-BA 2.0 mg/L KT 0.5 mg/L IBA 0.5 mg/L,and bud seedling differentiation ratio was 41.5% on this optimized medium.Moreover,the effects of different genotype and pre-cultivation on hypocotyls regeneration of Glycyrrhiza were investigated.The results showed that the hypocotyls regeneration rate of Glycyrrhiza inflate Bat are 44.7%,which was higher than the other three varieties;Pre-cultivated hypocotyls on a suitable medium for 4 d could promote adventitious bud regeneration rate.A system of direct adventitious bud regeneration from hypocotyls of Glycyrrhiza was established,which provided an effective protocol for gene transformation and fast propagation of Glycyrrhiza.
