斑茅20S蛋白酶体α亚基5基因的克隆与序列分析

摘    要:进行了甘蔗属近缘野生种——斑茅抗逆性基因克隆的研究。以斑茅为材料,用RACE技术克隆了斑茅的20S蛋白酶体α亚基5基因,其编码区为711 bp,编码237个氨基酸,并于genbank分析其核苷酸和推导的氨基酸序列的同源性,用Clustalx version 1.8软件分析其系统进化关系。结果表明,物种间蛋白酶体进化保守,其核苷酸序列与水稻、大豆、剑叶藓属植物的相应基因同源性分别为91%、81%、80%;其氨基酸序列与水稻、大豆、剑叶藓属植物的同源性分别达97%、95%、86%,与人的同源性也高达69%。

Abstract:Erianthus arundinaceus is a related species of sugarcane but is more tolerant to drought, poor soil, virus and insects. The gene of 20S proteasome subunit alpha type 5 from Erianthus arundinaceus, obtained by RACE, contained an open reading frame of 711bp, encoding 237 amino acids. Alignment of its nucleotide sequence and its deduced amino acid sequence were analysed in Cenebank, and the phylogenesis of the deduced amino acid sequence was carried out by using the Clustalx version 1.8 program. The result indicated that the proteasome was conserved between species with its nucleotide sequence sharing a homology of 91% with that of Oryza Sativa, 81% with Glycine Max, 80 % with Physcomitrella patens, and its deduced amino acid sequence being 97 % homologous to that of Oryza saliva, 95 % to that of Glycine Max, 86% to that of Physcomitrella patens, and 69 % to that of the human.
