大豆—根瘤菌共生固氮乙炔还原活体测定方法的研究

摘    要:
本文选用克拉克结瘤与不结瘤等位基因系大豆为实验材料,对乙炔还原活体测定盆栽大豆共生固氮方法进行研究。测定了不同生育阶段盆栽大豆共生固氮量,并将乙炔还原活体测定方法与总氮差值法、乙炔还原离体测定方法和同位素稀释各方法相比较。经三年实验表明,乙炔还原活体测定共生固氮的方法是快速、灵敏、简便、可靠的。所测共生固氮值极显著,高于乙炔还原离体方法估测值;与同位素稀释各方法相比较,鼓粒以前,乙炔还原活体方法测定共生固氮值较高,鼓粒以后,活体方法估测值偏低。 乙炔还原活体测定共生固氮为短时间内筛选大豆——根瘤菌最佳共生体提供了简便、快速的方法。

Abstract:
Clack soybeans of noduleted and non-noduleted isolines were used for the study of acetylene reduction method in vivo to measure symbiotic N fixation in pots. The amount of symbiotic N fixation was measured by acetylene reduction method in vivo at various development stage. The results from acetylene reduction method in vivo have been compared with those from total nitrogen difference method, acetylene reduction method in vitro and different isotope dilution methods based on tracer. The results in three years indicated that the method of acetylene reduction in vivo was move fast, convenient, simple and reliable. The reading of symbiotic N fixation from acetylene reduction method in vivo were remarkably higher than those from the method in vitro.In comparison with isotope dilution methods, the readings of symbiotic N fixation measured by acetylene reduction in vivo were higher before seed filling, and lower affer seed filling.We suggest that the acetylene reduction method in vivo is simple and fast from screening best symbiotic system of soybeans and nodule bacteria in short time.
