喹胺醇与鸡血浆蛋白结合的初步研究

摘    要:采用平衡透析法结合反相高效液相色谱法(RP—HPLC)测定鸡血浆中喹胺醇的蛋白结合率。色谱条件为ZORBAX Eclipse XDB C18色谱柱(250mm×4.6mm,5μm),流动相为乙腈:水(40:60,v/v),流速1.0mL/min,检测波长352nm。结果显示,喹胺醇浓度为1.1、5.5、22.0μg/mL缓冲液中鸡血浆的蛋白结合率分别为84.41%、75.30%、66.96%;透析平衡时间约为30h。表明喹胺醇的鸡血浆蛋白结合率较高,药物浓度与血浆蛋白结合率之间呈反比关系。

Study on the integratation between Kui'anchun and plasma proteins of chicken

Abstract:The binding rate of Kui'anchun to plasma protein of chicken was determined by using the equilibrium dialysis and reversed- phase high performance liquid chromatography (RP- HPLC). Conditions of RP- HPLC were ZORBAX Eclipse XDB C18 of chromatographic column ( 250 mm × 4. 6 mm ,5μm) , the mobile phase of aeetonitrile - water (40/60, v/v) , the flow rale of 1.0 ml. · min^ -1 and the UV detecting wavelength of 352 nm. The protein - binding rates were 84.41% ,75.30% and 66.96% al conceutration of 1. 1 μg/mL, 5.5 μg/mL, and 22.0 μg/mL in buffer, respectively. The time of dialysis was about 30 hours. The result showed that the binding rate of Kui'anchun to plasma protein was high in chicken, while the protein - binding rate was decreased with the increasing of drugs concentration.
