离子交换树脂固定化果胶酶条件的优化研究

摘    要: 以离子交换树脂为载体、戊二醛为交联剂,对果胶酶进行固定化。研究了温度、pH值、时间、加酶量、戊二醛浓度、交联温度、交联时间对果胶酶固定化效果的影响。研究表明,最佳固定化条件为:温度40℃,pH 5.5,固定化6 h,加酶量0.75 mL,戊二醛交联浓度0.1%,交联温度4℃,交联时间4 h。在此条件下固定化果胶酶的酶活回收率达到80%以上。

Studies on the Immobilization of Pectinase by Ion-exchange Resin

Abstract:Pectinase was immobilized on ion-exchange resin by cross-linkage of glutaraldehyde. Effect of temperature, pH, time, amount of the enzyme, concentration of glutaraldehyde, crosslinking temperature and time were studied for the impact of pectinase’s immobilization. The optimum conditions for the immobilization were as follows : 40℃, pH 5.5, 6 h, 0.75 mL pectinase. The concentration of glutaraldehyde was 0.1%, crosslinking temperature and time were 4℃ and 4 h respectively. Under the optimal conditions, the activity recovery of immobilized pectinase can reach to more than 80%.
