虎舌红叶片不定梢诱导研究

摘    要:目的]建立虎舌红的高频再生体系,为利用转基因技术改良其品质奠定基础。方法]以虎舌红试管苗顶端叶片为试材进行组织培养和不定梢诱导试验。结果]外植体在9种培养基上均能诱导出愈伤组织,4号培养基的不定芽再生率最高,为45.80%,植株生长健壮;其次是7号培养基,其不定芽再生率为39.76%。从整体上看,不定芽的再生率都不高,最高的也未超过50.00%。综合考虑愈伤组织诱导和不定芽再生,适合虎舌红叶片不定芽再生的培养基为:1/2 MS+TDZ 2.0mg/L+IAA0.2~0.4mg/L+AgNO3 3.0~5.0mg/L。选健壮苗接种于1/2MS+IBA 0.3mg/L+NAA 0.5mg/L培养基上,20d后生根率可达90%,炼苗后再生植株的移栽成活率达80%以上。结论]该研究成功建立了虎舌红叶片的离体再生体系。
