摘    要:通过苹果逆转座子长末端重复序列(LTR)保守区域设计引物,对影响苹果IRAP反应的重要因素进行了研究,建立并优化了苹果IRAP分子标记技术体系。优化的苹果IRAP分子标记体系为:25μL反应体系中,模板DNA50ng、Mg2+2.5mmol·L-1、dNTPs0.2mmol·L-1、10×buffer2.5mmol·L-1、TaqDNA聚合酶1.0U、引物0.4μmol·L-1。该体系在所试苹果品种中均获得了较理想的扩增效果,并能鉴定部分芽变品种。

Abstract:IRAP primers were designed based on Long Terminal Repeat(LTR) reserve region of retrotransposon in apple.Several important reaction factors of IRAP were studied in order to establish and optimize IRAP system in apple.The final optimal system was established as follows:template DNA 50 ng,Mg2+ 2.5 mmol·L-1、dNTP 0.2 mmol·L-1,10×buffer 2.5 mmol· L-1,Taq DNA polymerase 1.0 U,primer 0.4 μmol·L-1 in 25 μL reaction system.Ideal amplification results were obtained by using such IRAP system in tested apple cultivars,especially in identifying the bud mutation.
