人工老化对红麻种子活力及基因组DNA的影响

摘    要:以3份红麻种子为材料,对人工加速老化后种子活力变化及基因组DNA的遗传完整性进行研究.结果表明:高温(44±1)℃、高湿(95%相对湿度)老化处理后,红麻种子的发芽势、发芽率、发芽指数和活力指数均随老化时间的延长而降低,活力下降过程不是等速率的,品种之间存在差异;利用筛选的23对RAPD引物对3份材料的对照和不同老化处理的DNA进行遗传完整性检测,83-20和台农1号的等位基因频率都是100%,未检测到差异;检测到辽55的等位基因频率为100.85%,存在差异,经t-测验差异不显著,表明人工老化对红麻种子基因组DNA的影响不显著,即老化对红麻种质遗传完整性无显著影响.

Abstract:The vigor changes and the genomic DNA integrity were studied among 3 kenaf(Hibiscus cannabinus L.)seeds by artificial accelerated aging.Results showed that germination potential,germination rate,germination index and vigor index of these seeds declining along with seeds aging time on the condition of high temperature(44±1)℃ and high humidity(95%RH),while the decreasing rate of vigor was dynamic and there were differences in 3 varieties.Through checking the genetic DNA integrity of the control and the different aging treatments of 3 materials by using selected 23 RAPD primers,the result showed that the allele frequencies of 83-20 and Tainong No.1 were 100% without difference and Liao55 was 100.85% with difference,but the result of t-test were no significant difference,which indicated that artificial aging did not significantly affect genomic DNA of kenaf seeds,namely,aging had no significant effect on genetic integrity of kenaf germplasm.
