摘    要:本试验旨在研究白藜芦醇(Res)对氧化应激仔猪肠道黏膜结构、抗氧化能力、紧密连接蛋白及炎性因子mRNA表达的影响。选择健康状况良好、胎次相近的28日龄三元杂交(杜×长×大)断奶仔猪30头,随机分为5组,对照组饲喂基础饲粮,试验组在基础饲粮中分别添加0、10、30、90 mg/kg Res,每组6个重复,每个重复1头仔猪。于试验第15天清晨给试验组敌草快(Diquat)组、Diquat+10 mg/kg Res组、Diquat+30 mg/kg Res组、Diquat+90 mg/kg Res组]仔猪按照10 mg/kg BW的剂量腹腔注射Diquat建立氧化应激模型,对照组只注射相同剂量灭菌生理盐水为参照。试验期21 d。结果表明:与对照组相比,Diquat组绒毛高度和绒毛高度/隐窝深度(V/C)显著降低(P<0.05),隐窝深度显著升高(P<0.05);肠黏膜丙二醛(MDA)含量显著升高(P<0.05),谷胱甘肽过氧化物酶(GSH-Px)和超氧化物歧化酶(SOD)活性显著降低(P<0.05);肠黏膜白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)含量显著升高(P<0.05),而肠黏膜白细胞介素-10(IL-10)含量显著降低(P<0.05);肠黏膜闭锁小带蛋白-1(ZO-1)、闭锁蛋白(Occludin)和跨膜蛋白(Claudin)的mRNA相对表达量均显著降低(P<0.05)。与Diquat组相比,Diquat+30 mg/kg Res组和Diquat+90 mg/kg Res组回肠绒毛高度和V/C显著提高(P<0.05);肠黏膜MDA含量显著下降(P<0.05),GSH-Px和SOD活性显著提高(P<0.05),其中以Diquat+90 mg/kg Res组效果最为显著;肠黏膜ZO-1、Occludin和Claudin的mRNA相对表达量均显著提高(P<0.05),且不同Res剂量组间无显著差异(P>0.05);肠黏膜IL-1β、TNF-α和IL-6含量显著降低(P<0.05),同时IL-10含量显著提高(P<0.05)。综上所述,Res可以提高氧化应激仔猪回肠抗氧化酶活性,改善回肠黏膜形态结构,同时抑制肠道炎症的发生,维持肠道健康。

Abstract:The purpose of this experiment was to study resveratrol(Res)on the intestinal mucosal structure,antioxidant capacity,tight junction protein and inflammatory factor mRNA expression in oxidative stress piglets.Thirty 28-day-old three-way cross(Duroc×Landrace×Yorkshire)weaned piglets with good health and similar parity were selected and randomly divided into 5 groups,with 6 replicates in each group and 1 piglet in each replicate.The control group was fed a basal diet,and the experimental groups were intraperitoneally injected with Diquat+Res of 0,10,30 and 90 mg/kg were added to the basal diet,respectively.In the early morning of the 15th day of the experiment,the piglets in the experimental groups(Diquat group,Diquat+10 mg/kg Res group,Diquat+30 mg/kg Res group and Diquat+90 mg/kg Res group)were intraperitoneally injected with Diquat at a dose of 10 mg/kg BW to establish an oxidative stress model,and the control group only injected the same dose of sterilized saline as a reference.The trial period was 21 days.The results showed that:compared with the control group,the villus height and the ratio of villus height/crypt depth(V/C)of the Diquat group were significantly reduced(P<0.05),and the crypt depth was significantly increased(P<0.05);the content of malondialdehyde(MDA)increased significantly(P<0.05),the activity of glutathione peroxidase(GSH-Px)and superoxide dismutase(SOD)were significantly reduced in intestinal mucosa(P<0.05),intestinal mucosal interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)contents were significantly increased(P<0.05),while the intestinal mucosal interleukin-10(IL-10)content was significantly decreased(P<0.05);the relative expression levels of zonules atresia protein-1(ZO-1),Occludin and transmembrane protein(Claudin)in the intestinal mucosa were significantly decreased(P<0.05).Compared with the Diquat group,the ileal villi height and V/C in the Diquat+30 mg/kg Res group and Diquat+90 mg/kg Res group were significantly increased(P<0.05);the MDA content of the intestinal mucosa was significantly decreased(P<0.05),GSH-Px and SOD activities were significantly increased(P<0.05),among which the Diquat+90 mg/kg Res group had the most significant effect;intestinal mucosal ZO-1,Occludin and Claudin mRNA relative expression levels were all significantly increased(P<0.05),and there was no significant difference among different Res dose groups(P>0.05);intestinal mucosal IL-1β,TNF-αand IL-6 contents were significantly reduced(P<0.05),while IL-10 content was significantly increased(P<0.05).In summary,Res can increase the activity of antioxidant enzymes in the ileum of oxidative stress piglets,improve the morphological structure of the ileal mucosa,inhibit the occurrence of intestinal inflammation and maintain intestinal health.
