玉米赤霉烯酮中毒大鼠卵巢组织Bax和Bcl-2的表达

摘    要:10周龄SD大鼠单剂量腹腔注射5mg/kg玉米赤霉烯酮玉米油溶液,分别于3、6、12、24、48h时剖杀取卵巢组织,检测不同时间卵巢组织Bax和Bcl-2的表达。结果显示,病理组织学观察发现卵巢组织出现不同程度损伤,卵泡颗粒细胞发生凋亡。免疫组化SP法试验组与对照组卵巢组织中均有Bax和Bcl-2的表达,并且随着时间的推进呈现动态变化。Bax在3、6、12h时表达量上调,表达量与对照组比较差异显著(P〈0.05),24、48h时表达量下降,与对照组比较差异不显著(P〉O.05)。3~48h卵巢组织中Bcl-2表达量与对照组比较差异不显著(P〉0.05)。结果表明,大鼠玉米赤霉烯酮中毒可引起卵巢组织的病变及颗粒细胞凋亡,且Bax在玉米赤霉烯酮中毒大鼠卵巢颗粒细胞凋亡中起着重要作用,Bcl-2的作用不明显。

Expression of Bax and Bcl-2 mycotoxicosis of ovaries in SD rats exposured zearalenone

Abstract:Ten-week-old SD rats were treated with a single dosage intraperitoneal injection of zearalenone (ZEA) (5 mg/kg), and then the rats were euthanized to isolate the ovary tissues at 3, 6, 12, 24 and 48 h after treatment, respectively. Immunohistochemistry method was used to determine the expression of Bax and Bcl-2 in ovary tissues of rats. Histopathological observation found that ovarian tissues have different degree damages and apoptosis cells were observed in granular layer of ovarian follicle. SP analysis showed that the Bcl-2 and Bax were expressed, and presented dynamic changes as time went on in both groups and control group. The expression of Bax in the groups of 3, 6 and 12 h after treat ment with ZEA showed a significantly increase compared with the control group (P〈0.05), while the expression decreased by degrees after treated with ZEA in the groups of 24 and 48 h. The expression of Bcl-2 showed there was no statistical difference compared with control group. ZEA mycotoxicosis can result in the damage of ovarian follicles and apoptosis of granulosa cells in rats. Bcl-2 and Bax play an important role in apoptosis of granulosa cells in the rats with ZEA mycotoxicosis.
