3个不同品种羊Ⅰ型内根鞘角蛋白基因的差异表达

探讨了敖汉细毛羊、小尾寒羊和莱芜黑山羊3个不同品种毛囊生长期与毛囊发育有关的特异基因表达模式。采用基因表达谱芯片技术对上述3个品种羊毛生长期颈部、腹股沟部的Ⅰ型内根鞘角蛋白基因表达进行检测,通过实时定量PCR技术对差异基因进行验证。结果：Ⅰ型内根鞘角蛋白基因家族KRT25、KRT26、KRT27和KRT28在颈部的表达量无品种间显著变化（差异倍数小于2）;而在腹股沟部的表达量在品种间差异极显著（差异倍数大于2,P〈0.01）。实时定量PCR所得结果与基因芯片结果基本吻合。结果表明：Ⅰ型内根鞘角蛋白基因家族在不同品种羊中的表达量与特异部位毛囊分布密度有着重要的关系。

Differential expression of type I IRS keratin genes in three breeds of sheep

The study investigated the differential expression of specific genes related to hair follicle development dur- ing the growth and anagen phases among Aohan fine wool sheep, small-tail Han sheep, and Laiwu black goat. Expression of type I IRS keratin genes in the neck and groin during wool growth phase was examined by gene expression microarray technology in the three breeds. There were not significant differences （fold change 〈2） in the neck for the expressions of KRT25,KRT26 and KRT27 genes among the three breeds,but highly significant differences were found in the groins （fold change 〉2, P〈0.01）. Moreover,it is relative consistent with the results of geae expression microarray and real-time PCR. Expression of type I IRS keratin genes in different breeds of sheep was related to the hair follicle density of specific parts of the body.
