反硝化除磷系统的驯化及反硝化聚磷菌的筛选

摘    要:利用城市河道底泥,通过厌氧/缺氧/好氧(A2O)工艺驯化反硝化除磷系统,采用BTB培养基、异染粒及PHB(聚β-羟基丁酸)染色等方法,从反硝化除磷系统中分离筛选出反硝化聚磷菌,并通过16SrDNA序列测定分析其遗传背景。在该试验条件下,反硝化除磷系统的氮磷的去除率超过80%。从反硝化除磷系统中分离筛选出DPB—A511、DPB—A9和DPB—AIO3株反硝化聚磷茵,其氮、磷去除率均超过50%。这3株菌中DPB—A511、DPB—A9分别与Dechloromonas aromatica、Candidatus accumulibacter phosphatis的相似性均达到97%,DPB—AIO与Bacilluspumilus的相似性达到99%。

Abstract:Using the urban river sediments,denitrifying phosphorus removal system was successfully acclimated by anaerobic/ anoxic/ aerobic(A2O) process.Based on the system,denitrifying phosphorus accumulation bacteria were screened with BTB,metachromatic granules and PHB(poly-β-hydroxybutyrate) staining experiments,and their genetic backgrounds were found out by 16S rDNA sequence analysis technology.Under the experimental conditions,denitrification phosphorus removal system had more than 80% removal efficiency of nitrogen and phosphorus,and three denitrifying phosphorus bacteria were isolated from the system,which were named DPB-A511,DPB-A9 and DPB-A10 separately and had more than 50% nitrogen and phosphorus removal efficiency.DPB-A511 and DPB-A9 had 97% similarity with Dechloromonas aromatica,Candidatus accumulibacter phosphatis,and DPB-A10 had 99% similanty with Bacillus pumilus.
