光质对小苍兰茎尖试管培养的影响

摘    要:以小苍兰茎尖为外植体,分别置于白光(400~690nm)、红光(590~690nm)、黄光(520~640nm)、蓝光(400~530nm)、绿光(500~590nm)和黑暗下培养,研究不同光质对小苍兰茎尖试管培养的影响。结果表明:白光下愈伤组织及芽的分化速度最快,芽的分化率和生物量最高,试管苗叶片数多、生长良好,但愈伤组织诱导率及根分化受到明显的抑制;红光下根的分化速度最快,根分化率及综合培养力最高;蓝光对愈伤组织诱导率、叶绿素含量以及试管苗的干重/生物量有明显的促进;绿光和黑暗对茎尖的分化与生长均有不利影响,试管苗出现玻璃化现象。

Abstract:The explants from Freesia refracta meristem were cultured in white,red, yellow, blue,green light and dark respective,by studying the effects of light quality on meristem of its test tube culture. It was found that callus indution,organ differetiation,contents of chlorophylla and dry weight/biomass were greatly influenced by light quality. The results showed that under white light,speed of callus induction and bud differetiation were fast,differetiation rate of bud and biomass were hightest,leave of test tube seedlings were most,and seedlings grown very well. But induction rate of callus and differentiation rate of root were restrained obviously by white light. The speed of root differentiation was fast,differetiation rate of root and raising ability were highest under red light. Blue light could promote the rate of callus induction,contents of chlorophylla and dry weight/biomass of testtube seedlings notablely. Green light and dark did negative effects on differetiation and growth of meristem. The vitrification phenomena was found widespreadly in green light and dark.
