摘    要: 根据保守氨基酸序列FTQQYQ 和anti-1/M IWPNV 设计苹果自交不亲和基因引物P1 : 5′2 TT2TACGCAGCAATATCAG23′; P2 : 5′2 ACGTTCGGCCAAATA /CATT23′, 利用PCR - RFLP分析方法, 以河北省新引进的苹果品种美国8号、昂林、松本锦、华冠、南方脆幼叶为试验材料, 鉴定其自交不亲和基因型分别为S9S27、S1S9、S9S 9、S2S9、S2S9

Abstract:Five new apple cultivars,USA No.8,Korin,Matsumoto Nishiki,Huaguan and Nanfangcui,which were introduced into Hebei Province recently,were used as materials in the present study.The primers(P1:5'-TTTACGCAGCAATATCAG-3';P2:5'-ACGTTCGGCCAAATA/CATT-3') were newly designed according to the conserved sequence of amino acid of S-RNase.The self-incompatibility alleles of the cultivars were isolated and identified using PCR-RFLP analysis.Based on the results,the self-incompatibility genotypes of the cultivars were identified as S9S27,S1S9,S9S9,S2S9 and S2S9,respectively.
