番茄子叶离体培养与再生成株

摘    要:取番茄不同苗龄的无菌苗子叶进行培养,发现以14天苗龄的子叶培养不定芽再生频率最高。在MS培养基上添加不同浓度的吲哚乙酸(IAA)和玉米麦(Zt),对子叶愈伤组织的形成影响不大,但对不定芽的分化有较大影响,其中以MS+Zt1mg.L^-^1IAA1mg.L^-^1效果最好。在同样的培养基上继代培养,待芽长至1-2cm长时转到生根培养基上即可在短期内长出根系,成为再生植株。

Abstract:The cotyledons of axenic seedlings of tomato(Lycopersicon esculentum Mill.)were cultured in vitro.Using 14-day old cotyledons with petioles as explants and a combination of Zeatin and IAA in MS basal media,about 90% of the explants produced shoots in 2 weeks.The optimal conditions for regeneration were found to include a zeatin concentration of 1 mg.L-1 and IAA concentration of 1 mg.L-1.The roots developed from shoots.in the rooting medium and the regenerants were transferred into pots for further growth.
