摘 要: | 以白梨 (PyrusbretschneideriRehd )鸭梨的芽变大鸭梨试管苗为试材 ,试管苗幼叶为外植体 ,研究了培养基的基本元素组成、外源激素及接种叶片的放置方向对叶片不定梢再生的影响。结果表明 ,培养基的基本元素组成是影响叶片能否获得再生不定梢成功的关键因素 ;外源激素直接影响叶片的不定梢再生频率 ;叶片的接种放置方向影响单位叶片的再生不定梢数。诱导大鸭梨叶片获得高频率不定梢再生的最佳培养基为基本培养基NN6 9附加激素TDZ和IBA。叶片远轴面接触培养基 ,平均单位叶片产生的不定梢数量最多。

High frequency shoot regeneration from Giant Duckpear leaves in vitro |

Abstract: | Tube seedlings of Giant Duckpear which was a sport of Duckpear( Pyrus bretschneideri Rehd ) were selected as materials, with young expanded leaves extracted as explants Effect of element composition of basic media, foreign hormone and leaf orientation inoculated on media on shoot regeneration was studied The results showed that the element composition of basic media was the major factor affecting shoot regeneration Foreign hormones directly affected shoot regeneration frequency Leaf orientation inoculated on media affected average shoot number per leaf The optimal medium of inducing high frequency shoot regeneration was NN69 supplemented with TDZ and IBA Average shoot number per leaf was hightest when the leaf abaxial side touched media |
