高浓度CO_2饲养环境对柞蚕3龄幼虫部分生理指标的影响

摘    要:以柞蚕3龄幼虫为材料,在人工控制的高浓度CO2环境中进行饲养试验,调查幼虫的摄食量、生命力及血淋巴保护酶和中肠蛋白酶活力的变化,探讨高浓度CO2饲养环境对柞蚕幼虫生长发育及保护酶和蛋白酶活性的影响。结果表明,柞蚕3龄幼虫在高浓度CO2(5 000μL/L)条件下饲养,随饲养时间延长其摄食量与对照组相比呈减少趋势,死亡率呈升高趋势,摄食量在处理72 h后低于对照0.2 g/头,差异达到极显著水平;幼虫血淋巴中的过氧化氢酶(CAT)、过氧化物酶(POD)、超氧化物歧化酶(SOD)活性都低于对照组,其中CAT在处理24 h后与对照组的差异达到显著水平,处理72、96 h后差异均达到极显著水平,SOD在处理144 h后与对照组的差异达到显著水平;幼虫中肠中的总蛋白酶、强碱性类胰蛋白酶、弱碱性类胰蛋白酶活性都低于对照组,但没有显著性差异,类胰凝乳蛋白酶活性在处理24 h后极显著低于对照组。试验结果显示高浓度CO2饲养环境对柞蚕幼虫的生长发育及保护酶和蛋白酶活性均产生了影响。

Abstract:The influence of carbon dioxide(CO2) enriched atmosphere on growth and development of Antheraea pernyi larvae and activity of protective enzymes and proteases was investigated using the 3rd instar larvae of A.pernyi reared under artificially controlled CO2 enriched atmosphere.The food intake and vitality of the larvae were recorded,and the activity of protective enzymes in hemolymph and proteases in midgut was determined.It was found that larval food intake of the treatment group declined and mortality increased under enriched CO2 atmosphere(5 000 μL/L) compared with the control group.The average daily food intake per larva of the treatment group at 72 h was 0.2 g lower than that of the control group.This difference reached the extremely significant level.The activity of catalase(CAT),peroxidase(POD) and superoxide dismutase(SOD) in larval hemolymph from the treatment group was also lower than those of the control group,among which the difference of CAT reached significant level at 24 h and extremely significant level at 72 h and 96 h,and the difference of SOD reached significant level at 144 h.The activity of total proteases,strong alkaline trypsin-like enzymes and weak alkaline trypsin-like enzymes in larval midgut from the treatment group was all lower than those of the control group,but the difference did not reach significant level.The activity of chymotrypsin-like enzymes was extremely significantly lower than that of the control group at 24 h.These results demonstrate that CO2 enriched atmosphere had influence to the growth and development of A.pernyi as well as to the activity of protective enzymes and proteases.
