桦褐孔菌胞外多糖脱蛋白工艺比较研究

摘    要:采用正交试验设计,以蛋白质脱除率和多糖损失率为指标,比较研究了Sevag法、三氯乙酸法和酶法对桦褐孔菌胞外多糖的脱蛋白工艺。结果表明:三氯乙酸法脱蛋白效果最佳,其优化工艺为三氯乙酸浓度0.5mol/L,加入量15%,振荡时间20min,静止时间40min,蛋白质脱除率为68.5%,多糖损失率为2.45%。

Abstract:To investigate the optimal deproteinization methods from exopolysaccharide of Inonotus obliquus, Sevag, trichloracetic acid and enzymic method were compared according to the deproteinization and loss rate of exo polysaccharide by orthogonal experiments. The results showed that  the effectiveness of trichloracetic acid method was the best for deproteinization  of I. obliquus exo polysaccharide.The optimal conditions of trichloracetic acid method were as follows: concentration of trichloracetic acid 0.5 mol/L, volume of trichloracetic acid 15%; vibration time 20 min, quiescing time 40 min,respectively. The deproteinization ratio and loss rate of exo polysaccharide were 68.5% and 2.45%, respectively.
