摘    要:利用RAPD标记对17个结球甘蓝品种进行了分析。结果显示较低浓度基因组DNA、高浓度Taq酶及一致的Mg^2 浓度,有利于RAPD结果的稳定。所有品种经12个引物扩增后,发现利用其中4个引物在17个品种间所扩增出的多态性标记可将全部品种鉴别。17个品种的遗传多样性分析显示,秋冬甘蓝品种内的遗传差异较春甘蓝品种内更大。因此,为保持结球甘蓝多样性,将更多的遗传资源用于春甘蓝品种选育是必要的。

Abstract:Seventeen cabbage varieties were studied by means of RAPD markers in this research. It was shown that low concentration of genomic DNA, high concentration Tag DNA polymerase and similar Mg 2+ concentration in PCR reaction were beneficial for the stability of the results of RAPD. Twelve primers were used in DNA amplification, and 4 of them were found to show abundant polymorphism, sufficient to distinguish the 17 varieties. More intravarietal genetic diversity was observed in autumn-winter cabbage varieties than in spring varieties, suggesting that more germplasms should be used in cabbage breeding so as to maintain the diversity of the whole species of Brassica oleracea.
