摘    要:
分别采用S.Wilkie的CTAB提取方法及在此基础上进行了改良的CTAB法,以硅胶干燥、标本干燥和新鲜的平邑甜茶叶片为材料,提取出平邑甜茶基因组DNA,并进行DNA质量的检测。结果表明,利用改良的CTAB法能够获得较高质量的DNA,新鲜叶片中提取的DNA量最多,经过硅胶干燥的叶片提取的DNA比标本干燥的叶片多。

Abstract:
Two different CTAB methods were used to isolate the DNA of Malus hupehensis(Pamp.) Rehd var.mengshanensis Y.N.Li ex G.Z.Qian.One is the method reported by S.Wilkie and the other was modified based on the former one.The materials involved in this experiment were dried with three different methods:silica gel dried leaves,fresh leaves without drying and leaves from herbarium specimens.The genomic DNA was isolated by using two methods and its quality was examined.The results showed that the modified CTAB method...
