罗布麻叶总黄酮类化合物的酶解-溶剂提取工艺研究

目的]研究了酶法提取罗布麻叶总黄酮的工艺,为罗布麻叶总黄酮的提取提供参考。方法]罗布麻叶先以纤维素酶酶解预处理后再用溶剂乙醇提取其中的总黄酮。分别考查了酶解各因素如酶解pH值、酶用量、酶解温度、酶解时间以及溶剂乙醇的浓度对罗布麻叶总黄酮得率的影响,确定了罗布麻叶总黄酮的最佳提取工艺条件。结果]罗布麻叶总黄酮的最佳提取工艺条件为:pH值5,酶用量3mg/g,酶解温度45℃,酶解时间2 h,乙醇浓度50%。该条件下总黄酮得率达到4.6%。结论]以纤维素酶酶解预处理后再用溶剂乙醇提取罗布麻叶中的总黄酮得率明显提高。

Study on Enzyme-solvent Extraction Process of Total Flavonoids in Apocynum venetum L.

Objective] The objective of this study was to study the enzyme-solvent extraction process of total flavonoids in Apocynum venetum L.,and provide references for the extraction process of total flavonoids in Apocynum venetum L..Method] The Apocynum venetum L.was pretreated with enzyme,then the total flavonoids was extracted by solvent ethanol.The pH value,amount of enzyme,enzymatic hydrolysis temperature,enzymatic hydrolysis time,concentration of solvent alcohol was experimented.Result] The optimum technological conditions of extracting total flavonoids in Apocynum venetum L.were as follow:pH was 5,amount of enzyme was 3 mg/g,enzymatic hydrolysis temperature was 45 ℃,enzymatic hydrolysis time was 2 hours,concentration of alcohol was 50%.The yield of total flavonoids was 4.6% on optimum technological conditions.Conclusion] After pretreated with cellulase enzyme the yield of the total flavonoids was obvious increased.
