甲基胆蒽诱发小鼠宫颈鳞癌间质中肥大细胞的分布及Alcian蓝·藏红染色特征

摘    要:本文用甲苯胺蓝(TB)染色法和Alician蓝·藏红(AB—S)染色法研究了甲基胆蒽诱发的小鼠宫颈鳞癌间质中肥大细胞的分布和组化特征。结果显示,癌旁间质和深层间质中肥大细胞数量较多,颗粒致密。癌巢之间的癌间质中肥大细胞数量较少(p<0.05),颗粒稀疏,似为功能活跃的脱颗粒肥大细胞。宫颈鳞癌间质肥大细胞对Alcian蓝,藏红的亲和力随Alcian蓝染色时间的不同而变化。Alcian蓝染色时间延长,Aician蓝阳性肥大细胞所占百分比增加,本实验结果表明,Alcian蓝阳性或藏红阳性并不能说明肥大细胞颗粒中含有生物胺还是肝素。影响肥大细胞颗粒中糖胺多糖和Alcian蓝和藏红结合的因素可能是颗粒中的蛋白质,并非肝素或生物胺

Abstract:The distribution and histochemical characteristics of mast cells in the stroma of squamous cell carcinoma of the mouse cervix induced by 20—methylcholanthrene were investigated using differect staining methods of Aldan Blue—Safranin (AB—S) and Toluidine Blue (TB). It was found that the number of mast cells in the stroma around the tumor and in the deep stroma was significantly higher than that found within the tumor. The mast cells within the tumor were less granulated than those existed at the periphery of the tumor. The phenomenon suggested that the mast cells within the tumor were in a status of active degranulation. The affinity of mast cells for Aldan Blue and Safranin varied with the difference in Aldan Blue Staining time. The percentage of AB—positive mast cell, when stained for 4 days with Alcian Blue, were much more than that stained for 30 minutes. Thus, it seems that AB—S stainability of mast cells could not prove whether substances in mast cell granules are heparin or biological amines. It might be the proteins that affect the binding of dyes with mast cell glycosaminoglycans rather than heparin or biological amines.
