摘    要:目的]为解淀粉芽孢杆菌的应用提供科学依据。方法]从最佳碳源、氮源、初始pH值、发酵时间、发酵温度、接菌量和装液量的确定出发,针对一株新发现的解淀粉芽孢杆菌SAB-1进行发酵培养基的设计及发酵条件的优化。结果]从8种不同的碳源和氮源中确定了长效碳源水溶性淀粉、长效氮源棉籽粉与速效碳源葡萄糖、速效氮源酵母浸膏。并优化了基础发酵培养基的发酵条件:温度31℃、转速180 r/min、发酵时间26 h、初始pH值6.0、接菌量1%、装液量50 ml/250 ml。该条件下发酵后菌体产量为46.0×109/ml。结论]该解淀粉芽孢杆菌在设计发酵条件下基本达到了工业规模化生产的要求。

Research on the Design of the Medium for the Amyloliquefaciens Bacillus Fermentation and Optimization of Its Fermentation Condition

Abstract:Objective] The scientific basis of the application of amyloliquefaciens bacillus was provided through the research.Method] The design of the fermentation medium for a newly discovering amyloliquefaciens bacillus-SAB-1 was conducted for the optimal fermentation condition including the carbon and nitrogen source,the initial pH,the fermentation time and temperature,the inoculation amount and the amount of liquid in fermentation tank.Results] From 8 different kinds of carbon and nitrogen sources,the long-term acting carbon source-soluble starch,the long-term acting nitrogen source-cottonseed meal,and the available carbon source-glucose,the available nitrogen source-yeast extracting solution were selected for its culturing.And the optimized fermentation condition in basal medium was as follows: the temperature,87.8℉;the speed,180 r/min;the fermentation time,26 hours;the initial pH value,6.0;the inoculation rate,1% and the liquid volume,50 ml/250 ml,under which condition,the fermentation yield of the bacillus strain was 46.0×109/ml.Conclusion] The amyloliquefaciens bacillus fermentation in the design of condition reached basically to the demand of the scale industrial production.
