大豆色拉油中转基因成分检测技术研究

摘    要:针对大豆色拉油中DNA较难提取的问题提出了充分乳化、延长醇沉淀时间和反复富集小片段DNA等措施,有效地从大豆色拉油中提取到DNA,并通过大豆特异内源基因扩增得以证明。从两个不同品种商品大豆色拉油中检测出35S启动子和NOS终止子外源调控序列,并通过增加所用DNA模板用量检测出目的基因EPSPS序列,建立了大豆色拉油中转基因成分检测方法。

Abstract:DNA was efficiently isolated from soybean oil through several measures of efficient emulsification, elongating the time of ethanol precipitation and repeat concentration of small DNA fragments. It was proved that DNA was isolated successfully by doing PCR of soybean native gene lectin; 35S promotor and nos terminator were detected in two varieties of soybean oil; Target gene of EPSPS (herbicide resistant gene) was successfully detected by increasing the amount of DNA in PCR; Detection method of transgenic factors in soybean oil was established.
