苦马豆果实中总黄酮的提取和含量测定

摘    要:[目的]探索苦马豆果实中总黄酮的提取方法和含量测定方法。[方法]以采集于青海省贵德县的苦马豆果实为材料,运用微波技术提取该果实中的黄酮类物质,并以芦丁为对照品,采用分光光度法测定提取物中的黄酮类物质的总含量。[结果]采用微波技术从苦马豆果实中提取出黄酮类物质,对其进行全波长扫描操作,样品在波长510nm处显示最大吸收,吸光度为0.242,计算出苦马豆果实中总黄酮含量为2.17%。5次平行测定的精密度相对偏差RSD为1.14%。样品液稳定性实验表明,吸光度值在5—45min内基本不变。回收率实验表明,测定平均回收率为98.86%,RSD为2.73%。[结论]采用微波技术可提高总黄酮的提取效率。采用分光光度法的测定方法简便、快速、灵敏,结果准确可靠,可作为苦马豆果实中总黄酮的含量测定方法。
