葫芦白粉病病原菌观察及系统进化分析

摘    要:为确定侵染葫芦使其感染白粉病的病原菌,并鉴定其生理小种,通过种植葫芦材料,采集自然发病的葫芦病叶及分生孢子,显微观察感病叶片上白粉病菌的形态特征,克隆病原菌的ITS区并测序进行分析,在形态和分子水平证明单囊壳白粉菌(Podosphaera xanthii)是侵染葫芦使其感染白粉病的病原菌之一。通过进行与不同地区、不同寄主作物的白粉病菌ITS区的系统进化分析,进一步说明该病原菌为单囊壳白粉菌。并且通过种植13份葫芦科白粉病鉴别寄主,发现该生理小种为1号生理小种。

Abstract:In order to make sure the bottle gourd infected with powdery mildew for identifying the species and the physiological race of the pathogen in this study, naturally infected gourd leaves and conidia on them were collected to microscopically observe the morphological characteristics of powdery mildew pathogen on susceptible leaves, and sequence analysis of the cloned ITS region of the pathogen.It was proved that Podosphaera xanthiiwas one of the pathogens caused powdery mildew infection at the morphological and molecular level.After the phylogenetic analysis of ITS regions of powdery mildew pathogen in different regions and different hosts,the results showed that the pathogen was Podosphaera xanthii.We found that physiological race was physiological race 1 after identification of hosts of 13 Cucurbitaceae powdery mildew.
