茄子叶中茄尼醇的RP-HPLC分析

摘    要:目的]建立测定茄子叶中茄尼醇的RP-HPLC分析方法。方法]采用Agilent 1100 HPLC,Hypersil ODS色谱柱(250 mm×4.0 mm,5μm),流动相V(甲醇)∶V(乙醇)为60∶40等度洗脱,洗脱流速1.0 ml/min,柱温30℃,检测波长210 nm,保留时间约10.720 min。结果]茄尼醇质量浓度在6.90~220.80μg/ml(R=0.999 5)范围内线性关系良好,平均加样回收率(n=9)为99.94%,RSD=2.08%。通过与传统提取方法比较,确定最佳的提取方法为微波辐射法。结论]所建方法具有简便、快速、准确、重复性好的特点,适用于茄子叶中茄尼醇的含量测定。

Abstract:Objective] The study aimed to establish a sensitive RP-HPLC method for determining the content of solanesol in eggplant leaves.Method] The Agilent 1100 HPLC system,Agilent Hypersil ODS column(A I.D.250 mm×4.0 mm,particle size 5 μm) was operated at a column temperature of 30 ℃.The mobile phase was V(methanol)∶V(ethanol)=60∶40 at a flow rate of 1.0 ml/min.210 nm was set as the UV detection wavelength for solanesol,retention time was probable 10.720 min.Result] The results showed a good linear relationship within 6.90-220.80 μg/ml(R=0.999 5,the average recovery(n=9)is 99.94%,RSD=2.08%.Comparison with conventional extraction method,the optimum extraction method was microwave irradiation.Conclusion] The method was rapid,simple,accurate and good reproducibility,it was suitable for the determination of solanesol of eggplant leaves.
