石斛叶片蛋白质双向电泳技术体系的建立及优化

摘    要:以石斛叶片为材料,建立了一套适用于石斛蛋白质组分析的双向电泳体系,探讨了石斛叶片蛋白的提取、蛋白定量以及双向电泳的一些关键技术。结果表明,通过三氯乙酸(TCA)法提取、高压等电聚焦和两步平衡等方法可以获得稳定、清晰的双向电泳图谱,获得了可分辨蛋白质斑点数约613个。

Abstract:A 2-D(two-dimensional electrophoresis)protocol suitable for protein analysis in dendrobium leaf was set up.The protein extraction and protein excretion.as well as some key technologies of 2-D electrophoresis were studied.The resultsshowed that stable and clear 2D maps with about 613 protein spots could be obtained through the methods of extraction with trichloroacetic acid,isoelectric focusing under high-voltage and two-step equilibrium.
