摘    要:以三倍体毛白杨叶片为外植体,筛选适宜遗传转化叶片的诱导分化培养基和生根培养基,确定转化体最佳选择压浓度和抗生素浓度。结果表明:叶片诱导分化培养基以MS+6-BA 0.6 mg/L+NAA 0.1 mg/L附加1.5%蔗糖、0.6%琼脂效果最好,最佳生根培养基为1/2MS基本培养基附加IBA 0.4 mg/L、1.5%蔗糖、0.6%琼脂。选择压浓度以50 mg/L卡那霉素对叶片分化和嫩茎生根效果显著。培养基中附加300 mg/L的羧卞青霉素不仅有效的抑制农杆菌的生长,而且还对分化起促进作用。

Abstract:This study is done by taking triploid Chinese white poplar laminae as explant to serve as the culture medium for the inducement of buds and rhizogenesis which adapt to genetic transformation to make sure the best choice of the selected pressure density and antibiotic density to transformant.The result showed that the most suitable medium for the inducement of buds was MS 6-BA 0.6 mg/L NAA 0.1 mg/L,add 1.5% sucrose and 0.6% agar,and the best medium for rooting was: 1/2MS IBA 0.4 mg/L,add 1.5% sucrose and 0.6% agar.The selected pressure was: 50 mg/L kanamycin is prominent for inducement of buds and rhizogenesis,and culture medium with 300 mg/L Carbenicillin can not only inhibite A.tumefaciens growing,but also promote bud growth.
